葛台明,男,1966年1月出生,湖北武汉市人。博士、教授。第九届湖北省政治协商会议科技界委员,中国遗传学会、中国法医学会会员,湖北省生物工程学会理事,中国合格评定国家认可委员会(CNAL)实验室评审员、检查机构评审员。历任中国刑事科学技术协会特聘研究员,湖北警官学院客聘教授,公安部批青年专家人才库成员,湖北省法医学会物证委员会主任。

主要研究兴趣
本人早期的研究兴趣主要集中在植物细胞工程及植物细胞遗传学领域,尤其是植物细胞工程领域。特别地,在植物体细胞杂交研究方面,首次在小麦中建立了一套完整的电融合技术体系,并第一个获得了“小麦+多花黑麦草”的亚族间体细胞杂种及其成熟植株。调入新葡萄88805官网(武汉)后,该领域的兴趣主要集中在利用植物细胞工程技术进行植物次生代谢产物的工厂化生产、植(作)物的品种改良、脱毒复壮,以及转基因等方面。
调入新葡萄88805官网(武汉)后,本人将主要研究精力同时也放在了分子生态学及保育遗传学领域。利用新葡萄88805官网(武汉)生物地质与环境地质国家重点实验室、湿地演化与生态恢复湖北省重点实验室等研究平台和技术优势,我们重点研究了微卫星标记的开发技术,并在该领域取得了重要进展,尤其在具有长重复基元(motif)的SSR标记开发方面取得了重要突破。结合新葡萄88805官网(武汉)的学科优势和特色,今后的主要研究兴趣同时也将拓展至分子生态学、保护生物学(保育遗传学)、植物传粉生物学、群体遗传学、海洋生物学等学科。
总的来说,我的研究领域包括如下三个大的方面:
1、分子生态学及保育遗传学,包括利用SSR标记等分子生物学手段,研究动植物种群的群体遗传结构、濒危珍稀动植物遗传多样性及其保护生物学(保育遗传学)、植物传粉生物学、海洋生物分子生态及保育等方面。
2、植物细胞工程研究,主要从事利用植物细胞工程及相关技术手段,研究植物次生代谢产物的代谢机理及应用开发工作,同时也包括利用植物体细胞杂交、原生质体培养、花药及小孢子培养、茎尖脱毒与快繁等技术,进行作物的改良、野生植物资源的开发利用等工作。
3、法医物证及相关分子生物学检验技术研究。

RESEARCH INTERESTS
My research interests include three main fields: Plant Cell Biology, Molecular Ecology and Human Identification (or other identification related to forensic analysis).
Cell Biology and Plant Cell Engineering
My earlier interests were Plant Cytogenetics and Plant Cell Engineering. Before I entered Huazhong Agricultural University as a postgraduate student, I analyzed karyotypes of some plants, so I am familiar with chromosomal analysis. When I was a PhD student, I developed the electrofusion techniques using the ‘donor-recipient’ model for the production of somatic hybrids/cybrids between common cultivated wheat (Triticum aestivum L.) and Italian ryegrass (Lolium multiflorum Lam.). Considering that the two fusion partners are phylogenetically remote and are sexually incompatible, and both size and osmotic pressure of the protoplasts of fusion partners are different, the electrofusion difficulty can be anticipated.
Now, the main interests of these fields are focus on the following aspects: Cell suspension culture and secondary metabolites production, micropropagation and virus-elimination, in vitro production of haploids, transgenic plant regeneration.
Molecular Biology and Molecular Ecology
Thanks to the support from the National Key Laboratory of Biogeology and Environmental Geology (BGEG), and the Hubei Key Laboratory of Wetland Evolution and Ecological Restoration, currently, our lab has developed a novel strategy for isolating SSR markers, especially for isolating those with longer repeating unit (motif), for example, the trinucleotide repeating SSRs [e.g. (TTG)n] and tetranucleotide repeating SSRs [e.g. (GATA)n].The SSR markers with longer repeating unit are much better than those with shorter motif, because when amplifying the microsatellites with longer repeating unit, they displayed much weaker stutter bands, and easily to be analyzed. So the further interests should be focusing on the following:
1) Molecular Ecology and Conservation Biology of some endangered or economically important species (including marine species), especially using SSR markers to investigate evolutionary, ecological, and behavioural processes in these species. We hope that our research can help the Government to formulate conservation management plans for those species.
2) Reproductive Biology. According to Charles Robert Darwin’s theory about the formation of biodiversity through natural selection (and artificial selection on domestic species), all life is engaged in a perpetual struggle for survival and reproduction. Different organisms use different Gender Strategies to pass their genes to their offspring. By using SSRs (and other markers) to explore the mating (pollination) behaviour, I am also interested in trying to understand why the distribution areas of some endangered species were shrunken even without obvious disturbance of human being, and/or to understand the human impact on the endangered species.
3) New molecular techniques. For example, to isolate SSR markers in some non-model organisms, and to investigate the population structure of these organisms.
Human Identification
Based on my forensic experience, I am also interested in both analyzing the genotyping errors and investigating the distribution of some rare genes in human population. I am pleased to supervise some students in such field, especially those who come from the forensic system, or who wish to work in the field.
My interests also extend to the archaeological field. When I worked in the Criminal Investigation Department of Public Security Bureau of Hubei Province, I successfully extracted the sperm DNA in a body buried in wet sands for over 24 days (To my knowledge, this is the only successful example that had buried for a so long period). I also extracted DNA from skeletons buried for years. Thanks to the support from BGEG and the Criminal Investigation Department of Public Security Bureau of Hubei Province, I am also interested in using degraded DNA recovered from archived and archaeological samples to investigate past populations, in particular the influence of environmental change and exploitation on population dynamics and the evolution of species.
